It has suffered major population declines, local extinctions and range fragmentation. Several projects have attempted to reintroduce captive-bred birds into parts of the former range in Europe, but with little success. Two captive stocks currently exist, one originating from Pakistan in 1968 and the other originating from Spain in 1982. This study compares the suitability of these captive stocks for specific reintroduction projects by using 11 microsatellite markers and mtDNA control region sequences to assess genetic differences between captive populations and wild birds from Spain and Greece. No significant population structure was found and all microsatellite alleles recorded in captive birds originating from Pakistan were also observed in the wild Spanish population. A higher diversity of alleles was observed in wild birds from Greece than from Spain, probably due to the effects of a strong bottleneck experienced in Spain in the 1970s. Compared with wild populations, both captive stocks have suffered a significant loss of diversity in microsatellites and mitochondrial DNA owing to founder effects and/or genetic drift, and therefore may not be well suited for release programmes. We recommend the development of a more diverse captive breeding programme based on birds taken from different areas of the range, in particular by supplementing the Spanish population with birds from North Africa. Our study shows the value of molecular tools in developing conservation programmes for threatened bird species and has implications for the design of recovery programmes. It is generally split into two different specific entities: Apodemus epimelas occurs on the Balkan Peninsula and A. mystacinus inhabits Asia Minor and the Middle East. This analysis, based on two mitochondrial regions (cytochrome b and the D-loop) and the interstitial retinol binding protein (IRBP) nuclear gene, confirms an important level of genetic divergence between the animals from these regions and their separation from each other at least 4.2-5.1 Mya, which is in favour of a distinct specific status. Finally, the broad-toothed field mice from southwestern Turkey appear to be closely related to the animals from Crete but highly distinct from the populations of the other Oriental regions. This supports a distinct subspecific level (A. m. rhodius) for the insular animals and also for those from south-western Turkey. From a biogeographical point of view, it can be assumed that either late Pliocene or early Pleistocene cooling led to the isolation of two main groups of A. mystacinus, one in the Balkan region and the other one in Turkey and the Near East (Syria and Israel). In this region, it is suggested that a more recent event appeared during the Quaternary period, isolating broad-toothed field mice in Crete and leading to the appearance of two well-differentiated genetic groups: one in Crete and south-western Turkey, and the other widespread in northern and eastern Turkey as well as in the Near East. © 2005 The Linnean Society of London. Different data sets not only led to different evolutionary scenarios but also to different systematic classifications of the taxon. The present study uses detailed anatomical data for two pyrgulinid taxa, the type species of the subfamily, Pyrgula annulata (Linnaeus, 1767), and the type species of the little known genus Dianella, D. thiesseana (Kobelt, 1878), as well as DNA sequencing data of three gene fragments from representatives of eight rissooidean families to A) infer the phylogenetic position of Pyrgulinae with emphasis on its relationships within the family Hydrobiidae, B) to study the degree of concordance between anatomy- and DNA-based phylogenies and C) to trace the evolution of anatomical characters along a multi-gene molecular phylogeny to find the anatomical characters that might be informative for future cladistic analyses. Both anatomical and molecular data sets indicate either a very close or even sister-group relationship of Pyrgulinae and Hydrobiinae. However, there are major conflicts between the two data sets on and above the family level. Notably, Hydrobiidae is not monophyletic in the anatomical analysis. The reconstruction of anatomical character evolution indicates that many of the characters on which the European hydrobioid taxonomy is primarily based upon are problematic. The inability to clearly separate some hydrobiids from other distinct families based on those characters might explain why until only a few years ago, "Hydrobiidae" was a collecting box for numerous rissooidean taxa (mostly species with shells small and lacking any characteristic features). The present study not only stresses the need for comprehensive molecular studies of rissooidean taxa, it also demonstrates that much of the problems surrounding anatomical analyses in rissooidean taxa are due to the lack of comprehensive data for many representatives. In order to aid future comparative anatomical studies and a better understanding of character evolution in the species-rich family Hydrobiidae, detailed anatomical descriptions for P. annulata and D. thiesseana are provided. We Studied nine Populations from Greece, assigned to three nominal species (L. euboicum, L. germanicum and L. beieri), and two from central Europe (L. germanicum and L. 17 hypnorum), applying RFLP analysis of two mtDNA segments (12S rDNA and 16S rDNA) amplified by PCR. The analysis revealed seven monomorphic and four polymorphic Populations, and 17 different haplotypes. each one present in one population only. Several restriction enzymes produced Population specific patterns, enabling, discrimination among the different Populations. The estimated net nucleotide divergence bet,ween Populations ranges from 1.06 to 16.52%. while the high N-ST value (0.96) indicates the existence of increased interpopulation genetic differentiation that can be attributed to the extreme isolation among Ligidium populations. The relationships between haplotypes and Populations are not congruent with their geographical distribution and the established taxonomy within the genus. This is consistent with known absence of patterns of allozymes and morphological variation. Euscorpius italicus is found almost exclusively in human habitations. Its sister species, E. naupliensis, exhibits much higher genetic diversity within southern Greece. We suggest that the natural populations of the thermophilic E. italicus underwent a bottleneck during the glaciations, and that its modem range could be a result of dispersal with humans. Several karyotypic forms have been previously described in populations of the vole species Microtus thomasi from Greece. In particular, the karyomorphs Microtus thomasi ‘thomasi’ and ‘atticus’ differ in X chromosome morphology, being acrocentric and subtelocentric, respectively. Furthermore, remarkable heterochromatin content variability has been described in sex chromosomes of both karyomorphs. Genomic DNA digestion with AluI allowed us to clone an 884 bp long repeated DNA sequence (Mth-Alu900) from the karyomorph M. thomasi ‘atticus’. This repeated DNA is AT rich and seems to be organized mainly as a dimer of the 884-bp unit, which presents three simple repeats (CAAAT, CAGAT and CAGAC) that constitute 80% of the total unit length. This repeated DNA is exclusive to M. thomasi, since it is absent from the genome of other studied Arvicolinae species. The chromosomal location of Mth-Alu900 was analyzed on M. thomasi ‘thomasi’ and M. thomasi ‘atticus’ karyomorphs, with different sex chromosome constitution. It was mainly located on the pericentromeric heterochromatin of most autosomes and X chromosomes on both karyomorphs. Results are also discussed in relation to karyotypic and sex chromosome variations in M. thomasi. To our knowledge, Mth-Alu900 constitutes a new – the third discovered so far – pericentromeric repeated DNA sequence described in Microtus species.
